Matforskningsinstituttet Nofima (the Norwegian Institute of Food, Fisheries and Aquaculture Research (NOFIMA)) develops and uses methods for analysis of ingredients in complex matrices such as food, feed, ingredients, residual raw materials and digested food. The analyses are a central part of our work as a business-oriented research institute that delivers internationally recognised research and solutions that give Norwegian industry and commerce a competitive advantage along the entire value chain. For the above-mentioned analyses, NOFIMA intends to purchase a Thermo Ion trap Orbitrap ID-X LC-MSn instrument together with a Vanquish Flex liquid chromatography system and three types of ion sources of Nerliens Meszansky AS. Orbitrap mass analyser is patented technology that is only marketed by Nerliens Meszansky in Norway. 10 day waiting period from the publication in Doffin/TED. Lot 1: Food products contain complex mixes of natural, chemical ingredients that contribute both to nutrition, health and taste. When food is affected by cultivation or farming conditions, storage and preparation, the ingredients may change. Especially in plant foods, there are thousands of ingredients that belong to extremely varied chemical structure classes, and with either a completely unknown chemical structure, or with a very similar structure or molecular weight, but possible different effects on health and taste. To meet our need within method development and analyses, NOFIMA intends to purchase a LC-MSn instrument for structure clarification and quantification of plant metabolites and other connections in food, ingredients and digested material, as well as metabolomics work, lipidomics work and peptidomics work. In order to identify ingredients in food and avoid ambiguous reporting, we need a mass spectrometer that can fragment molecules in multiple consecutive steps at the same time as giving high resolution mass information (at least R = 150 000 FWHM) for small molecules and fragments so that the element composition and structure can be determined with sufficient accuracy. The instrument must have different mass analysers that must be able to run individually and in combination. In addition, we need the possibility of using multiple different fragmentation techniques on all fragmentation levels, which can trigger different fragmentation of the molecules or fragments, and provide completive structural information. The instrument must have high and stable mass accuracy (Dm/z = 200; mass area m/z = 50-2000), which is a requirement for many of the connections that will be analysed. It is also the only instrument that has three different mass analysers and the possibility for both CID and HCD fragmentation techniques on all fragmentation levels. Ion trap Orbitrap ID-X has sufficient mass accuracy at low concentrations, which are kept stable through internal calibration, and the possibility of selective and sensitive quantification. Ion trap Orbitrap ID-X has the possibility of multiple different ion sources and takes up relatively little space. The accompanying software provides the possibility of automatic, efficient fragmentation, which is important to simplify our omics type analysis, plus access to internet-based databases, fragment search and fragmentation simulation. A liquid chromatography system Vanquish Flex with DAD detector will be ordered together with the instrument. To make the instrument usable for multiple intended applications, NOFIMA intends to purchase both an APCI and a MALDI ion source suited to the Ion trap-Orbitrap ID-X instrument. Ordering also includes software that provides the possibility of simplifying omics work flow and identification of unknown, automated access to internet-based databases and integrated structure search and fragmentation prediction.
