Description

Novo Nordisk Foundation Center for Biosustainability intends to purchase a High Resolution-Accurate Mass mass spectrometer (HRAM-MS) for analysis of complex samples for metabolomics and compound identification. Novo Nordisk Foundation Center for Biosustainability intends to purchase a High Resolution-Accurate Mass mass spectrometer (HRAM-MS) for analysis of complex samples for metabolomics and compound identification. The MS instrument must besides high precision also be fast scanning, as the samples contains several thousands of compounds, and the analysis time need to be as short as possible. The MS must be capable of producing very detailed information on every single compound, which is of outmost importance for determining the chemical structure and thereby the identification of the analysed compound. The MS needs to be coupled to a binary HPLC system and all the parts must be controlled by the same software. To obtain the best coverage the instruments must have two independent mass analysers, which can be operated in parallel. In metabolomics/compound identification, the ability to identify small molecules from their mass spectra is crucial. Very high mass resolution is required to limit the number of possible chemical formulas, and a minimum of 60 000 FWHM is required. The instrument requested has a resolution up to 500 000 FWHM. Furthermore, high-resolution fragmentation patterns are needed for structural elucidation. The current instrument provides two modes of fragmentation — collision induced dissociation (CID) and higher-energy collisional dissociation (HCD) — and the ability to fragment multiple times (MSn), which is not available on any other high resolution MS. MSn significantly enhances structure elucidation, for example, in the analysis of lipids, where MS3 is usually required to identify the unsaturation locations on the acyl chain(s). The system will be an extension of CfB’s existing platform for compound identification. With only a few qualified operators of this highly specialised equipment, it is important that the system is fully compatible with the existing systems, in order to reduce errors and enhance mastery. This further ensure that analytical methods can be moved between instruments without further development and optimisation. CfB finds that the above-mentioned specifications only can be covered by the Fusion ID-X from Thermo Fisher Scientific.
