An upgrade to the University’s existing 400 MHz NM Spectrometer is required to include a state-of-the-art InsightXpress MR system, to enable the unique advantages of NMR spectroscopy to be applied in ultra-fast reactions. The addition of this system makes it possible, for the first time, to monitor species formed during reactions that reach completion in the microsecond timescale permitting analysis across a wide temperature range (−150 to +150 °C). This unit contains a stopped-flow pump that utilises a precisely timed trigger pulse to bring about rapid mixing of reagents as they are introduced into the spectrometer. The fast flow rates (0.2—2 ml s−1), extremely low dead-time (< 150 ms) and low sample volumes (< 600 μL) facilitated by this system results in both rapid sample addition and rapid sample mixing, enabling NMR spectra of homogenised samples to be recorded almost immediately after mixing. This allows previously unattainable reaction rate and profiling data to be recorded. The InsightXpress MR system incorporates a Bruker Avance III NEO console, containing 2 channels that each have transmit and receive capabilities, providing immense flexibility in terms of instrument configuration and multi-channel operation for maximum 1D/2D experiment choice. This configuration will ensure optimal use of the NMR system and allow future expansion into other techniques and applications (e.g. solid-state, MRI imaging, biological reaction studies). Coupled with the latest SmartProbe technology, an observe broadband high-resolution probe for 5 mm sample diameter will greatly extend the range of direct and indirect observeable NMR nuclei; tuneable for any nucleus in the range from 31P-199Hg and 17O-109Ag. A key feature of this probe is the ability to observe 19F with 1H decoupling and to perform two-dimensional 1H/19F spectroscopy with superior multiple solvent suppression using presaturation or pulsed field gradients as required for samples from reaction control experiments. A significant advance on our existing technology within the ProSpect facility is an extended temperature control range (—150 °C to +150 °C), which will be possible with the SmartProbe, thereby enhancing opportunity to slow the rate of fast interconverting or reacting species (millisecond) to a level where they can be identified and quantified.
